Recommended Dosage: Avapritinib is administered orally on an empty stomach, at least 1 hour before or 2 hours after a meal. Do not make up for any missed dose within 8 hours of the very next scheduled dose. Do not repeat the dose if vomiting occurs after taking the dose, but continue with the next scheduled dose.
GIST Harboring PDGFRA Exon 18 Mutations: The recommended dosage is 300 mg orally once daily in patients with GIST. Continue treatment until there is evidence of disease progression or unacceptable toxicity.
Advanced Systemic Mastocytosis: The recommended dosage is 200 mg orally once daily in patients with AdvSM. Continue treatment until there is evidence of disease progression or unacceptable toxicity.
Indolent Systemic Mastocytosis: The recommended dosage is 25 mg orally once daily in patients with ISM.
Treatment with Avapritinib tablets may rarely cause bleeding in the brain. Patients should report life-threatening headaches, dizziness, confusion, or vision changes. Regular monitoring is required, particularly in those with bleeding risks. Quick medical attention is required in case symptoms suggest internal brain bleeding.
Patients using this therapeutic may experience confusion, memory loss, or difficulty concentrating. These effects may occur weeks after starting this treatment. Let your doctor know if you or your family notice unusual behavior or thinking problems. The dosage adjustments may help manage signs and symptoms.
Avapritinib therapy can make the skin more sensitive to sunlight. Patients need to use sunscreen, wear protective clothing, and avoid prolonged sun exposure. Sunburns may occur more easily. Even on cloudy days, take proper precautions to protect yourself throughout treatment.
This medicinal product may cause harm to an unborn baby if used during pregnancy. Women of childbearing potential should use effective birth control while on treatment and for some time after the final dose. Discuss all the pregnancy risks and contraception options with your healthcare provider before starting the treatment.
Breastfeeding is not recommended during treatment with Avapritinib 25 mg, 50 mg, 100 mg, 200 mg, or 300 mg tablets, and for at least 14 days after the final dose of Avapritinib. It is not known if the medicine passes into human breast milk and may cause harm to a nursing infant. Talk to your healthcare provider (HCP) about feeding options.
