Columvi is a prescription anticancer medicine and must be administered under the supervision of a qualified hematologist-oncologist, in a setting equipped to recognize and manage CRS.
The recommended dosage of Columvi (glofitamab) is administered by intravenous (IV) infusion using a step-up dosing schedule. Before starting treatment with Columvi, patients receive one 1,000 mg dose of obinutuzumab by IV infusion. This is given 7 days before the first Columvi dose.
The initial dosage of Columvi is gradually increased to help reduce the risk of cytokine release syndrome (CRS).

Each treatment cycle is 21 days. Columvi is administered only by IV infusion. The infusion duration and required monitoring depend on the dose and the patient's clinical condition.
Treatment continues for a maximum of 12 cycles, or until disease progression or unacceptable toxicity, whichever occurs first. Premedication (including a corticosteroid, antipyretic, and antihistamine) is required before every dose, and adequate hydration should be ensured. Patients are generally hospitalized during and for 24 hours after the first step-up dose, and may need to be hospitalized for later doses if they experienced CRS previously.
