KRAS G12C-mutated Metastatic Colorectal Cancer (mCRC): In combination with panitumumab, for the treatment of adult patients with KRAS G12C-mutated mCRC, who have received prior fluoropyrimidine-, oxaliplatin- and irinotecan-based chemotherapy.
KRAS is one of the most frequently mutated genes in human cancers, and KRAS G12C mutations occur in approximately 10–13% of patients with non-small cell lung cancer, particularly in adenocarcinoma histology and in patients with a smoking history. For decades, KRAS mutations were considered undruggable, but sotorasib represents a breakthrough therapeutic approach. By selectively binding to KRAS G12C-mutant proteins, sotorasib irreversibly traps the protein in its inactive GDP-bound state, preventing downstream signaling activation and cancer cell proliferation. This mechanism-based inhibition reduces tumor growth, induces apoptosis in cancer cells, and improves progression-free and overall survival in KRAS G12C-mutant NSCLC.
Lumakras / Lumykras is designed for patients with confirmed KRAS G12C mutations who have progressed on at least one prior systemic therapy (chemotherapy, immunotherapy, or targeted therapy). Molecular testing to confirm KRAS G12C mutation status is essential before initiating treatment.
It is a prescription anticancer medicine and must be used under the supervision of a qualified medical oncologist or pulmonologist experienced in targeted oncology.
Sotorasib may increase liver enzymes and rarely cause severe liver damage. Regular liver function tests are recommended, especially during the first three months. Seek immediate medical attention for jaundice, dark urine, pale stools, severe fatigue, or abdominal pain.
Sotorasib may cause heart failure or reduced left ventricular ejection fraction (LVEF). A baseline cardiac evaluation, such as an echocardiogram or MUGA scan, with periodic monitoring during treatment is recommended, particularly for patients with cardiovascular risk factors.
Rash is a common side effect of Sotorasib. Rare but serious skin reactions, including Stevens-Johnson Syndrome (SJS) and DRESS syndrome, have occurred. Report any new, severe, or worsening rash immediately for prompt medical evaluation and treatment.
Sotorasib may increase the risk of serious infections. Patients should promptly report symptoms such as fever, chills, persistent cough, sore throat, or other signs of infection so that appropriate medical evaluation and treatment can begin without delay.
Muscle pain, joint pain, myositis, and fasciitis have been reported during Sotorasib treatment. Severe symptoms may require temporary treatment interruption or permanent discontinuation. Inform your healthcare provider about any persistent pain, weakness, or stiffness.
Sotorasib may harm an unborn baby. Women of reproductive potential should use effective contraception during treatment and for at least one week after the final dose. Breastfeeding is not recommended while receiving this medicine.
