Vonjo (pacritinib) is a kinase inhibitor approved by the FDA under accelerated approval for the treatment of adults with intermediate or high-risk primary or secondary myelofibrosis (including post-polycythemia vera and post-essential thrombocythemia myelofibrosis) who have a platelet count below 50 x 10⁹/L.
Vonjo inhibits JAK2 and IRAK1 signaling pathways involved in the abnormal blood cell production characteristic of myelofibrosis. Unlike some other JAK inhibitors, Vonjo is specifically indicated for patients with significant thrombocytopenia (low platelet counts), a population often excluded from other myelofibrosis treatments.
Vonjo is intended for myelofibrosis patients with severe thrombocytopenia (platelet counts below 50 x 10⁹/L), a group with limited treatment options due to bleeding risk concerns with other therapies.
The recommended dose of Vonjo is 200 mg taken orally twice daily, with or without food. A reduced starting dose of 100 mg twice daily is used in patients with severe hepatic impairment.
Vonjo does not carry a formal FDA boxed warning. Unlike some other JAK inhibitors, Vonjo's current label does not carry the class-wide JAK-inhibitor boxed warning for MACE, thrombosis, malignancy, and mortality — these risks appear as standard (non-boxed) Warnings and Precautions. Patients and prescribers should be aware of the following important safety information.
Bleeding events have been reported. Vonjo should be avoided in patients with active bleeding, and treatment should be held before planned surgery.
Diarrhea is commonly reported and may require dose reduction or temporary interruption.
Vonjo should be avoided in patients with a baseline QTc greater than 480 msec, and electrolyte abnormalities should be corrected before starting treatment.
Major adverse cardiovascular events and thrombosis have been reported, particularly in patients who smoke or have other cardiovascular risk factors.

Vonjo is manufactured and marketed by Sobi, Inc. (Swedish Orphan Biovitrum). It is sourced through authorized distribution channels for patients seeking named-patient import into India.
Patients typically need a valid prescription from a treating hematologist confirming myelofibrosis diagnosis and platelet count. NPP's regulatory team assists in preparing and verifying the documentation needed for a compliant import.
As an FDA-approved therapy for myelofibrosis patients with severe thrombocytopenia, Vonjo is sourced through authorized channels and imported under GDP-compliant logistics to preserve product quality throughout transit.
NPP sources Vonjo directly from authorized pharmaceutical distributors to ensure product authenticity and full traceability. Every shipment is handled under Good Distribution Practice (GDP) conditions from origin to the patient's doorstep.
Vonjo should be stored at room temperature, below 30°C, in its original package to protect from light, and this condition is maintained throughout transit.
Once documentation and regulatory approvals are in place, delivery timelines are communicated transparently at the time of quotation, accounting for import clearance and last-mile logistics to the patient's city.
