Leritrelvir is an oral antiviral tablet for COVID-19, developed in China by Guangdong Raynovent Biotech together with Guangzhou Laboratory and the Guangzhou Institute of Respiratory Health. It is sold there as 乐睿灵®. Unlike most medicines of its type, it is taken on its own, without a ritonavir booster, and a course lasts five days.
China's National Medical Products Administration (NMPA) granted it conditional approval in March 2023, and the company announced in August 2026 that this had been converted to a regular (full) approval. It is approved for adults with:
It is a prescription infection medicine and should only be taken on a doctor's advice, ideally soon after symptoms begin.
The coronavirus relies on an enzyme called the 3CL (main) protease to cut its proteins into working parts before it can copy itself. Leritrelvir blocks that enzyme and so stops the virus from multiplying. It stays attached to the enzyme for longer than earlier medicines of this kind, which is why it does not need ritonavir to keep its blood level up. That also avoids the long list of drug interactions that come with ritonavir.
The phase 3 trial, published in eClinicalMedicine, enrolled 1,359 adults aged 18 to 75 with mild or moderate COVID-19 at 29 sites in China, all treated within 48 hours of their first symptoms. Almost all (98.5%) were vaccinated. People taking leritrelvir recovered from all 11 tracked symptoms about 20 hours sooner than those on placebo (median 251 vs 271 hours). Viral load also fell faster, with an extra 0.82 log10 drop by day 4. Nobody in either group was admitted to hospital, became severely ill or died, so the trial could not test whether it prevents serious illness. The company also reports that a large real-world study found a lower risk of severe illness, including in people over 65 and those with other health conditions.
In the phase 3 trial the dose was:
One box holds 30 tablets, which matches this full 5-day course. According to the company, no dose change is needed for older adults, for mild or moderate liver impairment, or for mild, moderate or severe kidney impairment. Follow the dose your doctor prescribes and the instructions in the pack leaflet.

Blood fats: the most common side effects in the trial were raised triglycerides (11.7% vs 10.4% on placebo) and raised blood lipids (8.8% vs 7.7%). None of these was serious.
Overall: side effects were reported in 46.5% of people on leritrelvir and 43.5% on placebo; 32.2% and 27.7% respectively were judged possibly related to treatment. Serious events were rare (0.15% vs 0.45%), and only 0.29% stopped treatment because of side effects.
